The antitumor activities of water extracts from Agaricus blazei Murill were examined in vitro and in vivo. In the direct cytotoxic effect on several tumor cells in vitro, the water extract from fruiting bodies of Agaricus blazei Murill (CPF), showed more cytotoxic, dose-dependent effect than others against all tumor cells tested. In the in vivo study using sarcoma-180 tumor implanted BALB/c mice model, the positive group receiving lentinan i.p. showed the highest inhibitory effect among five experimental groups (p<0.05). Among the groups receiving water extracts of Agaricus blazei Murill, the oral administration of 400 mg CPF significantly inhibited the growth of sarcoma-180 tumors (p<0.1). We further examined in vitro macrophage activation as a potential mechanism leading to antitumor action. The J774A.1 macrophage-like cells stimulated with CPF produced a large amount of H₂O₂ in dose-dependent manner, while production by a mixture of extracts from mycelia and fruiting bodies (CPMF) was not significant. On the other hand, tumor necrosis factor (TNF) production was highest in 500 μg/well of CPMF-treated cell culture, but all tested concentration of CPF did not show enhanced production of TNF. This enhancement of H₂O₂ and TNF production by water extracts from Agaricus blazei Murill may play a role in the host-defense mechanism against tumors.
