    Solid freeform fabrication (SFF) technology like multi-head deposition system (MHDS) were used to fabricate hybrid constructs by layer-by-layer deposition of polycaprolactone (PCL) and chondrocyte cell encapsulated alginate hydrogel. Cell dispensing conditions and optimum alginate concentration for maintaining cell viability were determined. In-vitro assays to determine glycosaminoglycans (GAGs), DNA and Collagen II content from different PCL/alginate gel based constructs were performed. 4% alginate gels containing transforming growth factor (TGF-β) showed higher extracellular matrix formation. The hybrid scaffolds of PCL-alginate gel were implanted in the dorsal subcutaneous spaces of the nude mice. Histology analysis of the implants revealed enhanced cartilage tissue formation in the PCL/ alginate gel (+TGF-β) hybrid scaffold. In conclusion, we present an innovative biohybrid scaffold for cartilage regeneration fabricated by advanced bioprinting technology.
