Effects of methanol extracts from Artemisa iwayomogi (AI), Ganoderma lucidum (GL), Hericium erinaceus (HE), G. lucidum cultivated with A. iwayomogi (GLA), and H. erinaceus cultivated with A. iwayomogi (HEA) on biotransformation of aflatoxin B1 into aflatoxin B1-8,9-epoxide (AFBO) were evaluated. HEA showed the most potent inhibitory effect on AFBO formation, whereas that of HE was lower, possibly due to the changes in chemical components of the culture broth after the addition of AI. HEA also had stronger anti-complementary activity than HE. These results indicate the cultivation of HE with AI enhanced cancer preventive activity and host defense system. Anti-complementary activity and inhibitory effect on the AFBO formation of GLA were lower than those of GL. Low anti-complementary activities should specify low inhibitory effects on AFBO formation. Anticarcinogenic studies using natural products have been focused on the inhibitory effects in suppressing the carcinogenic effects of aflatoxins (Fig. 1) (6). Due to its potential carcinogenicity to animals and birds, aflatoxin B1 (1, Fig. 1) poses the most threat to public health and international trade (7, 8). One of the carcinogenic mechanism of aflatoxins is the formation of a single initial DNA adduct by aflatoxin B1-8,9-epoxide (AFBO) (2, Fig. 1) with a guanyl N7 atom in the DNA sequence (9). The AFBO formation is primarily catalyzed by CYP450 3A4 in human (10). Therefore, cancer chemoprevention studies have been focused on finding phytochemicals to disrupt the epoxidation reaction of aflatoxin B1. In addition, many studies have shown that various polymers from mushrooms possess some immuno-modulation properties on the human complement system, such as anti-complementary and interferon-inducing activities, that play pivotal roles in the defense system (11-13). The anti-complementary activities of various mushrooms have been extensively investigated for their fruiting bodies, mycelium, and culture broth and was revealed to be related to anti-tumor actions of mushrooms (14). In our study, we investigated the effects of five methanol extracts of oriental medicinal sources on the formation of the 8,9-epoxide derivative from aflatoxin B1. We also show that anti-complementary effects of the five methanol extracts are proportionate to the inhibitory effects on the AFBO formation.
