Allergen-specific immunotherapy (AIT) is the only identified immune-modulating intervention for allergic diseases at present. The major mechanism of AIT remains controversial, but it is generally believed that AIT alters T-cell and B-cell responses, reduces the activity of effector cells of allergic inflammation such as eosinophils and mast cells, and leads to desensitization of allergen subjects. Regulatory T (Treg) and B cells (Breg) are thought to play major roles in generating these suppressive effects following repeated allergen administration. Conventional AIT using natural allergens extracts or recombinant allergens often suffer from drawbacks such as life-threatening anaphylaxis. The development of novel candidates for AIT is therefore focused on safer and more effective strategies. The molecular characteristic of an optimal agent for immunotherapy should have minimal IgE-binding and yet retaining the ability for induction of tolerance. The muscle protein tropomyosin is well documented as the major allergen in shrimp and other shellfish. We are currently investigating three possible strategies for AIT of tropomyosin hypersensitivity, namely hypoallergens, T cell epitope peptides and mimotopes. Hypoall ... 전체 초록 보기
