Perfluorooctanoic acid (PFOA) is a widely used industrial chemical that induces reproductive toxicities. To gain a better understanding of the mechanism underlying the PFOA-induced spermatogenesis impairment, we investigated the effects of PFOA on spermatogenesis and germ cell apoptosis. Ten-week-old Sprague- Dawley rats were treated with PFOA 10 ㎎/㎏/day for 1 day or 5 ㎎/㎏/day for 3 days orally. Rats were euthanized in 12 hrs and 1, 2, 3 and 7 days after treatment. The effects of PFOA on the testes were examined by determining apoptosis and the possible role for Fas and Fas ligand (Fas-L) in atrophy of the testes by histopathology, TUNEL assays and immunoblot. PFOA administration did not lead to significant changes in body and testicular weights. All doses of PFOA showed similar pathological changes in the testes. Degeneration and apoptosis of germ cells was evident in 12 hr of PFOA treatment and progressively increased in a time-dependent manner until 2 days and then slightly decreased at 3 and 7 days after PFOA treatment. Fas and Fas-L protein levels were increased. Collectively, these data suggest that impaired spermatogenesis after PFOA treatment is associated with an increase in germ cell apoptosis and Fas and Fas-L play a significant role in the initiation and/or progression of PFOA-induced testicular damage.
