Vascular Dementia(VaD) causes changes in the hippocampus that may lead to cognitive decline in rats. In rat animals with VaD, exercise increases hippocampal neurogenesis and serotonin. We investigated whether treadmill exercise would benefit rats that were sedentary until 19 months of age. Specifically, rats were housed with or without treadmill exercise and injected with bromodeoxyuridine or retrovirus to label newborn cells. Exercise rats showed increase of hippocampal neurogenesis and serotonin in raphe compare to the VaD-induced rats. The decline in neurogenesis in VaD-induced rats was reversed to 50% of Sham group levels by running. Moreover, fine morphology of new neurons did not differ between sham groups and VaD+EXE group, indicating that the initial maturation of newborn neurons was not affected by VaD+EXE. Thus, voluntary exercise ameliorates some of the deleterious morphological and decreased serotonin expression of rats with VaD.
