In order to investigate the toxicity of carbofuran and compensatory effects of phenobarbital sodium(PB) against carbofuran toxicity on hepatic tissues and cytochrome P-450 enzyme system of the rat, Rats were administered carbofuran only, PB only or combination of carbofuran and PB. Carbofuran cause severe injury in the hepatic tissue in a dose and, time-dependent manner and increased the level of cytochrome P-450 enzymes at 6 to 12 hrs after administration. PB cause no hepatic injury, but the level of cytochrome P-450 enzyme system was increased. Histopathological changes were observed in hepatic tissues at 12 hrs after combination treatment with carbofuran and PB , but returned to control level at 48 hrs. PAS-positive cells were increased at 12 hrs after carbofuran administration only and most hepatic cells in carbofuran and PB treated animals showed PAS-positive reaction, but hepatic cells were protected. Glycogen granules were increased at 12 hrs after carbofuran administration, After carbofuran and PB combination treatment in best carmine stain. The levels of cytochrome P-450 enzyme system were increased after carbofuran and PB combination treatment more than after either carbofuran or PB treatment. The results indicate that PB reduced carbofuran toxicity probably by elevating the level of cytochrome P-450 enzyme system.
