인삼뿌리썩음병균(Cylindrocarpon destructans) 후막 포자의 형성은 CD(Czapek’s dox broth)와 SNY(Spezieller Nahrstoffarmer yeast extract broth)배지에서 진탕 배양(20℃, 120 rpm)하면 양호하였다. 특히 CD배지에서 20℃로 30일간 진탕배양하면 5.99 log 후막포자/ml로 형성되었다. 후막포자의 발아율은 12.9~23.3%로 CD배지와 SNY배지에서 좋았으며 발아적온은 5~10℃였고, 최적 pH는 6이었다. 그러나 20℃ 이상에서는 전혀 발아하지 않았으며 pH 7.0 이상에서는 발아율이 급속히 저하되었다. 인삼추출물 3%, GA 10 ppm, IAA 10 ppm, NOVOZYM™234 20 ppm을 처리할 경우 대조구에 비해 발아율이 증가하였다. 특히 NOVOZYM™234 20 ppm 처리구는 발아율이 무처리에 비해 2배 증가하였다. CD배지(pH 5)에 NOVOZYM™234 20 ppm과 GA 10 ppm을 조합처리하고 5℃로 정치배양 할 때 후막포자의 발아율은 49.4%로 가장 높았다.
Chlamydospores of Cylindrocarpon destructans were produced abundantly in the shaking culture of the fungus in Czapek's dox broth (CD) and Spezieller Nahrstoffarmer yeast extract broth (SNY) media. Particularly, 5.99 log chlamydospores/ml were produced when the fungus was cultured in CD medium at 20℃ for 30 days. Germination ratio of chlamydospores was 12.9~23.3%, relatively high, in CD and SNY media, but none in potato dextrose broth medium. The optimum conditions for the germination of chlamydospores were 5~10℃ and pH 6. No germination of chlamydospores was observed above 20℃, and germination ratio was decreased rapidly above pH 7.0. Treatment of chlamydospores with 3% ginseng extract, 10 ppm of GA and IAA solution and 20 ppm of NOVOZYM™ effectively increased germination, particularly treatment of NOVOZYM™234 (20 ppm) increased germination 2 times compared with control. Germination ratio was greatest (49.4%) when chlamydospores were incubated at 5℃ in CD medium (pH 5.0) treated with NOVOZYM™234 and 10 ppm GA.
