Although applying herbal medicines by their precise discrimination is a crucial issue, numerous herbal medicines accommodate problems in reality starting from the confusion between identifying the true and false medicine to its distribution mixed with poor products. In addition, even though diverse oriental medicine discrimination methods have been actually utilized to this day, there is still a room for improvement. Therefore, this study aims to establish basic techniques for the fundamental studies of herbal medicine through development of a technique to discriminate herbal medicines by using their molecular-biological characteristics, furthermore identify the origins of respective herbal medicines, and create a genetic method actually applicable on the subjected herbal medicines sold in market places.
The subject medicines in this study were Cervi Parvum Cornu, Dipsaci Radix, Bupleuri Radix, Fritillaria thungergii Bulb., and Artemisiae iwayomogii Herba. As for the Cervi Parvum Cornu, its identification took place in form of a pilot study for verification of sensitivity using capillary electrophoresis method. In addition, absolute quantification method using real time-PCR was applied in order to establish an investigation method for discrimination using high throughput technique. As for Dipsaci Radix, evaluation on the relative affinity between Phlomis umbrosa Turcz and Dipsacus asperoides took place through RAPD method and analysis on ITS region. Discrimination of Bupleuri Radix from Bupleurum longiradiatum Turcz, which is a false product, also took place through evaluations on ITS and rbcL region, and furthermore, the plants in the same genus as Bupleuri Radix were classified. In order to select excellent Fritillaria thungergii Bulb. for plant variety fostering, as the distribution of which is mostly relying on import, fractional extraction from seven varieties took place and their electron donating ability and antibacterial effect were measured while their specific markers for respective varieties were sought for through RAPD method. As for Artemisiae iwayomogi, one of the Artemisiae plants difficult to discriminate in dried up states, its specific marker was sought for also through RAPD analysis.
