During the tuber dormancy, a number of physiological processes occur which result in changes in endogenous metabolites. However, little is known about the biochemical or molecular events occurring during dormancy maintenance or loss. Previous studies have indicated that the regulation of protein synthesis may play a role in the transition from a dormant to a non-dormant state. In this study, cDNA clones that were differentially expressed in dormant, non-dormant and non-sprouting, and sprouting in potato tubers were isolated using differential screening and characterization of expressed sequence tags. Many of the differentially expressed cDNAs shared sequence homology with other plant genes, while others were previously un characterized. A total 26 different groups of the clones were isolated: five of which were previously characterized in potato, 14 of these clones shared homology with genes isolated from other organisms, and seven clones did not share significant homology with characterized genes. Northern blot analysis of RNA from dormant, non-dormant/non-sprouting, and sprouting tubers indicated that many of the cDNA were differentially expressed during loss of tuber dormancy.
