Laser microdissection (LMD) is an increasing important method for obtaining pure cell samples for genetic and proteomic analysis. Previous reports suggested that the latent infection of canine herpesvirus (CHV) might be occurred in the neuronal cells of infected animals. The aim of the present study was to identify the latent cells for the CHV by using LMD as an alternative method for localizing viral distribution. The microdissection of target cells by LMD was performed with the paraffin-embedded tissues from 3 puppies inoculated with CHV, which were suspected as the status of latent infection. Then, CHV-specific polymerase chain reaction (PCR) and real-time PCR were conducted with the extracted DNAs from the microdissected cells. As the results, we represent the final demonstration that CHV DNAs can be detected in the trigeminal ganglia cells of CHV-infected puppies. These findings indicate that trigeminal ganglia cells are susceptible to CHV and play an important role of the latent stage in natural host. Also, in this study, LMD was shown its possibility of wide applications for sensitive localization of low copy viral sequences within specific phenotype cells to investigate the role of viruses in a variety of clinical conditions.
