Melatonin protects neuronal cells from ischemic brain injury. This study investigated whether melatonin modulates the heme oxygenase (HO)-1 and HO-2 in ischemic brain injury. Adult male rats were treated with melatonin (5 ㎎/㎏) prior to middle cerebral artery occlusion (MCAO). Brain tissues were collected at 24 hr after MCAO and infarct volumes were analyzed using triphenyltetrazolium chloride staining. This study confirms the reduction of infarct volume as well as the decrease of number of terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling positive cells by melatonin treatment. Expression levels of HO-1 and HO-2 in the cerebral cortex were measured using Western blot analysis. In the presence of melatonin, HO-1 levels were increased in the ipsilateral cortex, compared to that of vehicle-treated animals. The level of HO-1 was 0.85±0.08 and 1.12±0.11 in the ipsilateral cortex of vehicle- and melatonin-treated animals, respectively. However, the level of HO-2 was consistently maintained in the ipsilateral cortex of vehicle- and melatonin-treated animals. The results of this study provide further evidence that melatonin prevents cell death caused by ischemic brain injury and HO-1 activation by melatonin may contribute to the neuroprotective effects.
