    Recent studies have shown that stem cells improve neurological deficits when transplanted into animal models of neurological disorders. Neurotrophins such as brain-derived neurotrophic factor (BDNF), ciliary neurotrophic factor (CNTF), neurotrophin-3 (NT-3), and nerve growth factor (NGF) are important modulators of neuronal function and survival. Their expression in the CNS after various insults is thus of major therapeutic consequence. The goal of our current study was to investigate whether adipose tissue derived-stem cell (ADSC) transplantation affects the expression of neurotrophins in neurodegenerative lysosomal disease, Niemann-Pick Type C (NP-C) mice. Our findings showed high BDNF expression but not CNTF, NT-3 and NGF in the ADSC transplanted NP-C mice compared to sham transplanted control mice. These results strongly suggest that BDNF expression by ADSC transplantation prevents Purkinje neuron loss in the cerebellum of NP-C mice. In summary, the results in the present study demonstrated that the transplantation of ADSCs activates neurotrophin and thus that it might be useful as a therapeutic tool for treatment of neurodegenerative brain disease.
