The aging process in cardiac muscle is characterized by a decrease of resistance against oxidative stress, particularly in down-regulation of antioxidant enzymes. Recent evidence suggests that the oxidative and inflammatory signaling may be a target of aging. This study compares changes in the oxidative and inflammatory responses as well as expression of antioxidant enzymes in young (5 weeks) and aged (50 weeks) mice heart after exposure to treadmill exercise training for sixteen weeks. Here the author demonstrated that aging increased cyclooxygenase-2 (COX-2) and nuclear factor kappa B (NF-κB), while decreased expression of heme oxygenase 1 (HO-1) and NAD(P)H: quinone oxidoreductase 1 (NQO1) in heart muscle as well as levels of total glutathione (GSH) in erythrcyte. However, sixteen weeks of treadmill exercise training increased the expression of HO-1 and the level of total GSH, while markedly reducing NF-κB DNA binding activity and COX-2 expression in heart muscle of old mice. These results indicate that treadmill exercise training attenuates oxidative and inflammatory signaling and promotes antioxidant defenses in aged heart muscle.
