This study reports the method of pollen analysis to classify honey samples according to floral sources, in samples from Korea. The pollens in honey have different molecular weights depending upon the honey floral sources. We purified the pollen in honeys produced from mono-floral and multifloral honey, respectively. Honey samples used in this study differ due to plant sources, collection periods, and each of the honeys were centrifuged by sucrose density gradient. This study describes a simplified method for determining the total amount of pollen grains and the relative frequencies of pollen from various plant sources in honey. In many honey samples commercially obtained from Korea beekeepers the pollen of plant species were obtained from Prunus sargentii, Castanea crenata var. dulcis, Robinia pseudoacacia and premium grade Honey. The data reflects the pollen situation of the honey where a particular honey was produced and the identification of plant origin based on the presence of a combination of pollen types regarding that particular floral source. The pollens in honey have different genotypes upon the plant sources. Analysis of the pollen 26s rDNA result showed different amplification froms by PCR. The use of s sucrose density gradient and PCR combined with pollen identification proved useful in characterizing honey samples from different botanical origins.
