Fenfluramine (FEN) is an effective appetite suppressant known to produce its anorectic effects through stimulating serotonin (5-HT) release and acting as a potent selective serotonin reuptake inhibitor (SSRI). In rats, acute administration of FEN produces a rapid decrease in feeding levels that persists for hours. However, chronic administration of FEN results in a loss of anorectic potency within a short number of days despite continued drug administration. The loss of efficacy with regards to appetite following chronic FEN treatment is not only observed in rodents but in humans as well. However, the central mechanisms underlying this tolerance to FEN are still largely unknown. We explore the possibility that chronic FEN treatment leads to compensatory increases in the expression of other 5-HT transport mechanisms, which reduce the ability of FEN to maintain elevated 5-HT signaling.
