Oct4 is a member of the OCT protein family.The Oct-4gene encodes a transcription factor that isspecifically expressed in embryonic stem cells and germ cells ofthe mouse embryo. The key role of Oct4 in the pluripotency regulator network is well documented. The Oct4 has been shown as a master regulator in maintaining the pluripotecy and self?renewal in embryonic stem cells (ESCs), but it’s not essential for the initiation of totipotecy and pluripotecy. This conclusion is further supported by the formation ofOct4 -GFP ? and Nanog- expressing inner cellmasses (ICMs) in embryos with complete inactivation of both maternal and zygotic Oct4 express ion and thereprogramming of fibroblasts into fully pluripotent cells by Oct4 -deficient oocytes. In addition, Oct4 is also an important component in generating iPS cells. In contrast to its critical role in pluripotency, maternal Oct4 expression is not critical for developmental competence of the oocyte. In this review, we summarize the function of Oct4 in mammalian embryonic development.
