The vector pCW5 with plasmid pC7, originalyisolated in Lactobacillus paraplantarum C7 derived fromkimchi, was constructed using a p32 strong promoter, the pC7replicon, and green fluorescent protein (GFP) as the reporter.E. coli andLeuconostoc mesenteroides, and GFP expression detectedusing a Western blot analysis. GFP fluorescence was recognizedin E. coli and Leuconostoc mesenteroides using a confocalmicroscope. In adition, GFP fluorescence was also clearlydetected in several industrialy important lactic acid bacteria(LAB), including Lactobacillus bulgaricus, Lactobacillusparaplantarum, and Lactobacillus plantarum. Thus, pCW5was shown to be effective for when using GFP as the reporter, and it can also be used as abroad-host-range vector for other lactic acid bacteria.
