Griseofulvin has been used as an antifungalantibiotic for the treatment of mycotic diseases of humans andveterinary animals. The purpose of this work was to identify agriseofulvin-producing endophytic fungus from Abies holophyllaand evaluate its in vivo antifungal activity against plantpathogenic fungi. Based on nuclear ribosomal ITS1-5.8SITS2sequence analysis, the fungus was identified and labeledas Xylaria sp. F0010. Two antifungal substances werepurified from liquid cultures of Xylaria sp. F0010, and theirchemical identities were determined to be griseofulvin anddechlorogriseofulvin through mass and NMR spectral analyses.Compared to dechlorogriseofulvin, griseofulvin showed highin vivo and in vitro antifungal activity, and effectivelycontrolled the development of rice blast (Magnaporthe grisea),rice sheath blight (Corticium sasaki), wheat leaf rust (Pucciniarecondita), and barley powdery mildew (Blumeria graminis f.sp. hordei), at doses of 50 to 150 μg/ml, depending on thedisease. This is the first report on the production of griseofulvinand dechlorogriseofulvin by Xylaria species.
