Plant regeneration protocols via callus induction from seeds of Miscanthus sinensis, a promising bioenergy crop, were developed. Basal media and 2,4-D concentrations, proline concentrations, and kinds and concentrations of gelling agent were tested to find out the optimal conditions for callus induction from mature seeds. On the combination treatment of basal media,MS medium and N6 medium, and 2,4-D concentrations, callus induction rate was the highest (42.2%) in MS medium with 4.0mg/L 2,4-D. Proline improved callus formation from seeds, and high concentration (12.0 g/L) of proline was good for callus induction. In gelling agent treatment, Phytagel was better for callus formation than agar and 2.25 g/L Phytagel produced the highest (86.1%) callus induction. Calli obtained from seeds were subcultured every 4 weeks. After 3 months of callus induction,calli with dense yellow color and dome shape were selected and used for plant regeneration. The callus, cultured on hormone-free MS medium or MS medium with 1.0 mg/L BA, produced adventitious shoot. The regenerated shoots were elongated and rooted on MS medium. The plantlets were acclimatized successfully and the regenerated plants showed normal phenotypes.
