Recombinant inbred lines (RILs) are preferable in genetic mapping because each RIL is fixed for different parental alleles in various combinations of linkage blocks. RILs can be developed by successive selfing or sib-mating of individual F2 progeny until a high level of homozygosity is achieved. A set of 502 F4:7 RILs were developed from a cross between Eunhakong, a yellowseeded sprout soybean [Glycine max (L.) Merr.] and KLG10084, a wild soybean (G. soja) with green seed-coat. Single seed decent and pedigree methods were used to advance the F2 to F4 and F5 to F7 generations, respectively. Various agronomic characteristics were investigated for evaluation of RILs. Days to flowering, days to maturity, number of nodes, number of branches, number of pods per plant, plant height, seed weight, and grain yield per plant showed significant variation among the RILs. Seed coat colors of RILs divided three classes including 166 lines with yellow, 333 lines with green, and 3 lines with mixed colors. However, in hilum color only 17 lines had yellow, 338 lines in buff and brown , 129 lines in black, and 18 lines in gray color. The mean 100-seed weight was 5.9±1.2 g for F4:6 RILs and ranged from 3.1 to 10.3 g. The mean score of the 502 F4:6 RILs for viny growth habit was 3.5 (1-erect like a cultivar, 5-mid-type, 9-viny like a wild soybean). The close relationship in most RILs to the erect parent implies that many of these RILs could be used in a single cross to an adapted parent in the development of natto and sprout soybean varieties. The highest yielding F4:5 lines, MCS192, MCS234, and MCS459, showed about 87% seed yield of Eunhakong. MCS85 and MCS501 showed similar or slightly higher sprout yield than that of Eunhakong. Several soybean RILs with erect growth and <10 g per 100-seeds were selected for the use in breeding for improved sprout and natto cultivars. Therefore these RILs from a Glycine max × G. soja can be used in breeding for improved sprout and natto varieties and for QTL mapping of sprout related characters, seed composition, branching patterns, and plant shapes.
