Isoprenoids form one of the most diverse group of natural products. Biosynthesis of isoprenoids via the 1-deoxy-D-xylulose-5-phosphate (DXP) pathway requires equimolar glyceraldehyde 3 phosphate and pyruvate to divert carbon flux toward the products of interest. In this study, we demonstrated that precursor rebalancing is one of the critical steps for the biosynthesis of isoprenoids in Escherichia coli via the DXP pathway. First, we constructed a lycopene-overproducing E. coli strain by the amplification of the native DXP pathway using synthetic promoters and redesigned 5’-untranslated regions (5’-UTR).
