Lipid A and core O-antigen polysaccharide (OP) are the main components of lipopolysaccharide (LPS) in Gram-negative bacteria, which may play an important role in host immune responses. However, no suitable method has been developed for the preparation of single lipid A and core OP fractions to evaluate their effects. To solve this problem, we developed an efficient protocol for LPS extraction from an attenuated S.
