Cancer is a top target in medicine and a tremendous efforts and resources have been invested to find efficient and safe therapies. Cancer chemotherapy directly tackles a key pathogenic effector such as oncogenic proteins, with limited efficacy, poor specificity, and the development of resistance, altogether contributing to clinical shortcomings. In contrast, gene therapy targets a molecular cause of cancer such as oncogenes and is capable of genetically manipulate undruggable pathways, while its cost-effectiveness, extent of therapeutic effects, and availability for a broad range of cancers are remaining challenges.
