Oxidative stress induced by free radicals formed in the human body crucially contributes to the aging process. Among the compounds with known antioxidant effects, flavonoids are a group of well-known antioxidant phenolic compounds. The ethyl acetate-soluble fraction of the Epimedium koreanum Nakai (Berberidaceae) methanol extract exhibited the potent DPPH radical scavenging activity. From the ethyl acetate fraction, ikarisoside A was isolated by several chromatographic processes. Ikarisoside A was tested for the effects of superoxide dismutase (SOD), catalase, and oxidative stress resistance by using Caenorhabditis elegans along with the active oxygen species (ROS) level. To determine whether the regulation of stress response genes in the ikarisoside A-treated C. elegans was responsible for the increase in stress resistance, SOD-3 expression was measured using transgenic strain CF1553. As a result, the ikarisoside A increased the SOD and catalase activities, reduced the ROS accumulation in a dose-dependent manner, and furthermore, the CF1553 worms treated with the compound exhibited higher SOD-3::GFP intensity compared to control worms.
