Freeze-drying is widely used for microbial preservation. However, maintaining viability without cryoprotectants remains a challenge, particularly for Saccharomyces cerevisiae. This study aimed to optimize the freeze-drying parameters without using protective additives. Key variables, including pre-treatment temperature and duration, initial and final shelf temperatures, and initial cell concentration, were systematically evaluated. The highest viability was achieved with pretreatment at 4℃ for 24 h, an initial shelf temperature of -0.5℃, a final shelf temperature of 40℃, and an initial cell concentration of log 9 cells/mL. Under these conditions, the viable cell count reached 1.0 X1010 cells/g with a viability yield of 21.6%. These results show that appropriate adjustment of freeze-drying conditions can preserve yeast viability without the need for cryoprotectants. This approach offers a cost-effective alternative for industrial applications and provides a foundation for future improvements through strain-specific optimization or selective use of protective agents.
