Background: Selaginella tamariscina (P. Beauv.) Spring is an important medicinal plant known as ‘Gwonbaek’ in Korea. However, its morphological similarity to S. stauntoniana Spring and S. involvens (Sw.) Spring can lead to misidentification in herbal materials. Therefore, reliable molecular markers are needed to distinguish these species.
Methods and Results: Complete plastome sequences of the three Selaginella species were compared to identify species-specific sequence variations. Phylogenetic analysis based on multiple-sequence alignments resolved the three species into distinct clades. Based on Insertion or Deletion (InDel) regions in the plastomes, four gel-based InDel markers were developed and evaluated using 10 Selaginella samples. The markers distinguished the three species based on their PCR amplification patterns and identified a market sample as S. tamariscina.
Conclusions: The four plastome-based InDel markers showed preliminary discriminatory ability among the tested samples of the three Selaginella species, demonstrating their potential for the molecular identification and quality control of Gwonbaek-related medicinal plant materials.
