This study evaluated the spatiotemporal distribution and species-specific acoustic standing stock of fishery resources in the Current Fishing Maintenance Area and the Korea–China Provisional Measures Zone of the Yellow Sea using fisheries acoustics and environmental DNA (eDNA) data collected in 2022 and 2023. Fish echoes were separated using 38 and 120 kHz data, and nautical area scattering coefficient (NASC) values were integrated at 1 nmi elementary distance sampling units. eDNA metabarcoding was used to identify major taxa, whereas eDNA read counts were not converted directly to biomass. Major taxa were selected by survey period, total NASC was partitioned among taxa, and species-specific target strength and length–weight information were applied for biomass conversion. Mean NASC in the Current Fishing Maintenance Area was higher in 2023 than in 2022 for April, August, and November, with 2023/2022 ratios of 3.59, 2.88, and 4.63, respectively. Species-specific standing stocks varied markedly among surveys; the largest 2023 estimates were 5,008.5 ton for Thunnus orientalis in April and 2,491.6 ton for Larimichthys polyactis in November. These results indicate that acoustic and eDNA data can provide complementary information on fish distribution, species composition, and standing stock in the Yellow Sea.
