Mitochondrial dysfunction is a common factor in metabolic and neurodegenerative diseases; bone pathologies; and cancer. Estrogens, in addition to their nuclear effects, modulate mitochondrial function by increasing energy production and regulating antioxidant balance. The selective accumulation of estrogens in the mitochondria is a strategy to concentrate their effects and enhance energy metabolism, which would represent an alternative to restore mitochondrial dysfunction. The lipophilic cation triphenylphosphonium (TPP) was coupled to two estrogens: 17β-estradiol, and 17α-ethinylestradiol, generating two novel mitochondria-targeted estrogens (mitoE2 and mitoEE2, respectively).
