Background: The genus Bupleurum (Apiaceae) includes the medicinal species B. komarovianum,B. longiradiatum, and B. latissimum, which are often misidentified or mixed in herbal markets. Therefore, reliable molecular markers are needed for accurate species discrimination and prevention of adulterations and misuses.
Methods and Results: Complete plastid genomes (plastomes) and 45S nuclear ribosomal DNA (nrDNA) sequences of B. longiradiatum and B. latissimum were newly assembled from Illumina MiSeq data using the de novo assembly of low-coverage whole-genome sequencing method and analyzed together with previously reported B. komarovianum sequences for comparative analysis. Further comparative analysis with six additional genomes from GenBank revealed species-specific single nucleotide polymorphisms. Based on these variants, we developed six single-nucleotide polymorphism (SNP) markers and validated them using high-resolution melting (HRM) analysis. The HRM profiles clearly distinguished the three species. Phylogenetic analysis using the maximum likelihood method supported species-level clustering, with B. latissimum forming a genetically distinct clade.
Conclusions: Complete plastomes and nrDNA sequences of three Bupleurum species were assembled, and SNP markers were developed and validated for species discrimination. These markers enable accurate authentication and help prevent adulteration in herbal market.
