This study reports the first isolation and polyphasic characterization of Aeromonas salmonicida subsp. salmonicida (ASS) from cultured Chinook salmon (Oncorhynchus tshawytscha) in South Korea. Diseased fish exhibited typical signs of furunculosis, including cutaneous and anal hemorrhage, splenic ulceration, and systemic inflammation across major organs. Five bacterial isolates were identified as typical ASS by 16S rRNA and vapA gene sequencing and clustered with ASS reference strains in the phylogenetic analysis. All 13 virulence- and metabolism-related genes tested (aerA, act, hlyA, exsA, carB, lon, gmhA, fur, rfbA, dctA, mglA, rpoS, and purD) were detected in isolates ASS-2 to ASS-5, whereas exsA was absent in ASS-1, indicating substantial pathogenic potential for host colonization and systemic infection. Broth microdilution testing revealed phenotypic variation in antimicrobial susceptibility; ASS-2, ASS-3, and ASS-4 were resistant to oxolinic acid (≥4 μg/mL) and oxytetracycline (≥8 μg/mL), whereas ASS- 1 remained susceptible to most agents tested. These findings confirm the natural occurrence of virulent, antimicrobial-resistant ASS in Chinook salmon aquaculture in South Korea, highlighting the need for active disease monitoring and prudent antimicrobial management.
