Based on 1 year-long randomized double-blind placebo-controlled clinical study at one of Korean reputed hospitals, the combined test material (Estromon<SUP>®</SUP>) of the new phytoestrogen FGF271 (the extract of the three mixed herbs), amino acid, vitamins and minerals has been significantly effective to improve such menopausal symptoms as vaginal dryness and hot flush, serum osteocalcin level and bone mineral density (BMD) of femoral bone. This in vitro research was done to find out the phytoestrogen effect of FGF271 while in vivo research was designed to investigate the sole effect of FGF271 by comparing with Estromon<SUP>®</SUP>. In result, cell proliferation and alkaline phosphatase (ALP) induction study of MCF-7, Ishikawa, and SaOS-2 cell shows that Cynanchum wilfordii, Phlomis umbrosa, Angelica gigas, and FGF271 have phytoestrogenic effect, respectively and FGF271 has higher phytoestrogenic effect than the individual herbal extract. In ovariectomized (OVX) animal experiment to investigate the sole effect of FGF271 by comparing it with Estromon<SUP>®</SUP>, the oral administration of 100 ㎎/㎏ of FGF271 per day for 12 weeks significantly increased BMD of femoral bone of rats compared to ovariectomised control group (P<0.05) while the group of 100 ㎎/㎏ of Estromon<SUP>®</SUP> did not show significant difference statistically. In conclusion, it was revealed that the improvement of menopausal symptoms and BMD of femoral bone in the clinical study was caused solely by the phytoestrogenic effect of FGF271 in the Estromon<SUP>®</SUP>.
