The purpose of this study was to investigate the effect of a dietary alternation and acute prolonged swimming exercise(3h;30min×3) on genes closely associated with fatty acid oxidation (β-HAD, FAT/CD36, PDK4, PGC-1) in various rat tissues(Vastus lateralis[Skeletal]; visceral adipose tissue[VAT]; brown adipose tissue[BAT]). Total 32 Sprague-Dawley rats were allocated one of two dietary conditions: either standard chow diet (CHOW, n=16) or high fat diet (FAT, n=16). Rats in each dietary condition were further divided into either exercise (n=8) or non-exercise group (n=8). In skeletal muscle, β-HAD gene expression showed statistical interaction between diet and exercise(p<.05). The expression of PGC-1 mRNA was shown no interaction between diet and exercise even though it was significantly increased following exercise(p<.01) in skeletal muscle. Although FAT/CD36 expression was shown a significant higher in FAT than CHOW(p<.05), there was no statistical interaction between diet and exercise in VAT. PGC-1 mRNA expression was significantly increased following diet and exercise(p<.05, p<.001), even though it was also shown interaction between diet and exercise(p<.05) in VAT. All genes were no shown statistical interaction between diet and exercise in BAT. In BAT, PDK4 gene expression was significantly increased in FAT after diet(p<.05) and PGC-1 was also significantly increased following exercise in exercise group(p<.05). β-HAD gene expression was significantly higher in VAT and BAT than skeletal(p<.001) and FAT/CD36 mRNA expression was also significantly higher in VAT than other tissue(p<.001). In BAT, PGC-1 expression was higher than other tissue(p<.05). In conclusion, this results was suggested that the expression of fatty acid regulatory genes may be modulated by independent mechanism on diet and exercise, respectively. And we indicated that gene expression was difference between skeletal and adipose tissues.
