Browning of the white adipose tissue is beneficial in treatment and prevention of the obesity and metabolic diseases. Exercise is known to induce browning genes in adipose tissues and improve insulin sensitivity. However, the browning effect of exercise depending on the sites and types of adipose tissue has not been determined. We investigated exercise-induced browning effects on different adipose tissues by analysis of histology and thermogenesis-related gene expression. Male and female CD1 mice were exercise with treadmill running with a speed of 12m/min at 10 degree incline for 30 minutes per day for 6 weeks. There was no significant difference in total body weight of male mice after 6 weeks of exercise, although significant decrease in body weight was observed in exercise groups for 2 and 4 weeks of female mice. The weights of the male genital and inguinal adipose tissues were significantly decreased after 2 weeks of treadmill training, whereas no significant difference was observed between the control and exercise groups of female mice. RT-PCR analysis revealed that expressions of UCP-1 and Cidea increased in both male genital and inguinal adipose tissues after exercise, with earlier onset of UCP-1 expression in genital adipose tissue. Hematoxylin-eosin stain and immunohistochemistry showed a decrease in the size of the lipid droplets in white adipocytes with increased number of capillaries after exercise, which was observed earlier in genital than inguinal adipose tissue. These results suggest that exercise of moderate intensity induces browning as early as 2 weeks with male and visceral fats more prone to browning by exercise.
