To investigate anticancer activity of ginseng, peptides were isolated from Panax ginseng C. A. Meyer, and analyzed for their anticancer activity. Water extract of ginseng powder was fractionated by hydrophobic XAD-2 column chromatography using 100% ethanol as an eluant. The hydrophobic fraction was found to show high anticancer activity on cell line (P388D1, a mouse lymphoma) with an IC₅₀ of 13.3 μg/mL. According to amino acid analysis and sequencing result, the anticancer peptides were composed of tryptophan with a sequence of either trp-trp or trp-trp-trp. The synthetic tripeptide, Trp-Trp-Trp, had the highest anticancer activity on P388D1 cell, whereas Trp in free amino acid form exhibited no effects on the cell. IC₅₀ values of dipeptide and tripeptide were 442.9 and 184.9 μg/mL, respectively. In the DNA cell cycle analysis of the tumor cell treated with the peptide fraction by flow cytometry, the DNA contents of S and G2/M phases in the cell cycle decreased, whereas that of G0/G1 phases increased.
