This study inferred phylogenetic relationship of 18 species of waders (Scolopacidae, Charadriidae, Laridae) and 2 species of Pica pica and Streptopelia orientalis as the out-group another order in Korea at the species level using COI (cytochrome c oxidase I) gene polymorphisms. Although many waders could be identified at the species level, the family Charadridae and Scolopacidae were only identified at the family level in this study. The Kimura 2 parameter (K2P) distances showed that the average intraspecific and interspecific distances were 0.44% and 16.1%, respectively. The average intra- and intergeneric distances were 3.89% and 16.54%, respectively. It indicates that gene substitution event took place much more robustly as the genus diverged and the event lessened after the divergence. A maximum likelihood (ML) tree was used to develop a monophyletic taxon supported with a bootstrap value of 50% or more. Our inter-genus grouping supports the current taxonomy. However, C. alexandrinus was divided into two groups. As a result of the analysis of 38 individuals in Charadrius. alexandrinus, haplotype 1, 2 were separated from the old continent and haplotype 3, 5, 7 and 8 from the new continent. These results may suggest the possibility that individuals from the new world are C. nivosus as a result of the median-Joining network analysis and high intra-species distance of 3.29. The phylogenetic analysis of waders in Korea is very useful for future study including the DNA barcode metadata of a large number of species, alongside the increasing activity of the global DNA barcode industry.
