Osteocalcin (OC) is the most abundant noncollagenousprotein of extracellular matrix in the bone. In an OC deficientmouse, bone formation rates are increased in cancellous andcortical bones. OC is known as a negative regulator of mineralapposition. OC is also expressed in the tooth of the rat, bovine,and human. However, little is known about OC during toothdevelopment in Xenopus. The purpose of this study is tocompare the expression of OC with mineralization in thedeveloping tooth of Xenopus, by using von Kossa stainingand in situ hybridization. At stage 56, the developmental stageof tooth germ corresponds to the cap stage, and an acellularzone was apparent between the dental papilla and the enamelorgan. From stage 57, calcium deposition was revealed byvon Kossa staining prior to OC expression, and thedifferentiated odontoblasts forming predentin were located atadjoining predentin. At stage 58, OC transcripts weredetected in the differentiated odontoblasts. At stage 66, OCmRNA was expressed in the odontoblasts, which was alignedin a single layer at the periphery of the pulp. These findingssuggest that OC may play a role in mineralization andodontogenesis of tooth development in Xenopus.
