Inducible HSP70s encoded by two almost identicalhsp 70 genes, known as hsp 70.1 and hsp 70.3 arelocated in a tandem array on mouse chromosome 17.Duplication of this gene has been found in varioushigher organisms. However, the role of two almostidentical hsp 70 genes is still unclear. To elucidate theambiguity of expression patern between hsp 70.1and hsp 70.3, we investigated the induction of eachhsp 70 isoforms by several HSP70-inducible stres-sors in wild type ( hsp 70.1+/+ and hsp 70.3+/+) and thehsp 70.1-/- (hsp 70.1-/- and hsp 70.3+/+) murine embryofibroblast (MEF) cells, and the M-1 mouse corticalcollecting duct cell line. Each induced hsp 70 iso-forms by heat shock were very similar at transcrip-tional and translational levels in wild type andhsp 70.1-/- MEF cells. The mRNA stabilities of bothhsp 70.1 and hsp 70.3, even in two kinds of hsp 70.3transcripts, were also very similar. L-azetidine-2-car-boxylic acid, sodium arsenite, CdCl2 and ZnCl 2caused induction of both isoforms of the hsp 70genes, even though their expression levels werevariable. NaCl caused induction of just hsp 70.1 geneexpression. H 2O2 failed to induce the expression ofeither hsp 70 genes in MEF cells, caused induction ofhsp 70.1 gene expression in the M-1 cell line. Hsp70accumulation by H 2O2 and NaCl treatment wasmainly due to hsp 70.1 expression. Our studies dem-onstrated that two hsp 70 genes respond differen-tially to types of stress.
