A maltogenic amylase gene from Lactobacillusgasseri ATCC33323 (LGMA) was expresed in Lactococcuslactis MG1363 using the P170 expresion system. Thesucesful production of recombinant LGMA (rLGMA) wasconfirmed by the catalytic activity of the enzyme in liquid andsolid media. The N-terminal amino acid sequencing analysisof the rLGMA showed that it was Met-Gln-Leu-Ala-Ala-Leu-, which was the same as that of genuine protein, meaningthe signal peptide was efficiently cleaved during secretion tothe extracellular milieu. The optimal reaction temperature andpH of rLGMA (55oC and pH 5, respectively) and enzymatichydrolysis patterns on various substrates (β-cyclodextrin, starch,and pullulan) supported that rLGMA was not only eficientlysecreted from the Lactoccus lactis MG1363 but was alsofunctionally active. Finally, the branched maltooligosaccharideswere effectively produced from liquefied corn starch, byLactoccus lactis, with a yieldof 53.1%.
