In order to investigate the possibility of using low-temperature aged mixed medicinal herb extract (Angelica gigas Nakai, Rechmannia glutinosa Libosch, Paeonia lactiflora Pall, Cnidium officinale Makino, Scutellaria baicalensis Georgi) as a material for beauty foods and herbal cosmetics, in the present study, we investigated the yield, total polyphenol content, antioxidant and NO production inhibitory effects of low-temperature aged mixed extracts. The mixed medicinal herbs was precipitated in mineral water and aged at low temperatures. After that, the supernatant was MLE and the precipitate was extracted with water (MLWE) and ethanol (MLEE). As a result, the yield of each extract was much higher in MLE (64.96%) than MLWE (8.30%) and MLEE (7.04%), the total polyphenol content was 283.9 mg TAE/g in MLEE, which was 2.2~3.8 times higher than that in MLE and MLWE. Antioxidation measurements showed that the radical scavenging abilities of DPPH (95.1%) and ABTS (96.7%) were significantly higher at 1,000 μg/mL concentration. In antimicrobial activity test, the MLEE showed a high antimicrobial activity against P. acnes. The cytotoxic concentrations of low temperature aged mixed medicinal herb extracts showed cell viability above 90% at concentrations 100 μg/mL and inhibition of NO production by 52.6% at 100 μg/mL for MLEE. From the results of this study, the ethanol extract of low temperature aged mixed medicinal herb showed a significant effect on the total polyphenol content, antioxidant, and inhibition of NO. Therefore, it is believed that the ethanol extract of low temperature aged mixed medicinal herb can be used as a material for beauty foods and herbal cosmetics.
